I'm pretty sure we could argue that anything against pre-marital sex (or shared bedrooms) has bedrocks in religion rather than common sense.
In the future, the Berlin wall will be a mile high, and made of steel. You too will be made to crawl, to lick children's blood from jackboots. There will be no creativity, only productivity. Instead of love there will be fear and distrust, instead of surrender there will be submission. Contact will be replaced with isolation, and joy with shame. Hope will cease to exist as a concept. The Earth will be covered with steel and concrete. There will be an electronic policeman in every head. Your children will be born in chains, live only to serve, and die in anguish and ignorance.
The universe we observe has precisely the properties we should expect if there is, at bottom, no design, no purpose, no evil, no good, nothing but blind, pitiless indifference.

Bedrock? I'll bet we could argue even more strongly for bedrocks in economics. And far more than bedrock for any location which doesn't have plentiful, convenient, and cheap contraceptives.
Last night as I lay in bed, looking up at the stars, I thought, “Where the hell is my ceiling?"

Oh poo with you and your convenient buxom ladies

Even so, given the current dispassion eugenics, argue that regulating sex in hotels is against this or that amendment?

If this happened in America, you mean? It's not a constitutional violation, no, but Congress would be able to regulate any attempts to deny service via the interstate commerce clause, as it did with the Civil Rights Acts, concerning denying service based on race. There is not presently a federal statute forbidding discrimination based on orientation or marital status. At the state level, some jurisdictions have such legislation and some do not. Congress would be unable to pass legislation mandating "no premarital sex in hotels" on the basis that privacy rights extend to control over our own bodies, but I don't think it is within the scope of federal power to require sex in hotels. And no one, particularly Nevada, wants to see what would happen if the Feds took it upon themselves to ensure that everyone in a hotel was having sex. Talk about your unfair government practices, using federal muscle to out-compete the nascent legalized sex trade.
